The Unstable Behavior of a Chromosomal Fragment of NICOTIANA PLUMBAGINIFOLIA Responsible for Chlorophyll Variegation in N. TABACUM.

چکیده
HE origin of chromosome instability and subsequent chlorophyll variegation Tin hybrid derivatives of Nicotiana tabacum (n=24) and N . plumbaginifolia (n=10) has been described by MOAV (1961) and GUPTA (1965, 1968b). It was demonstrated by (GUPTA 1968b) that the alien chromosome of N . plumbaginifolia (pbg) carrying a Ws gene (chlorophyll factor) showed instability in association with the N . tabacum (tbc) genome (homozygous for the white seedling loci, ws,, ws,, ws,, us,), and formed somatic anaphase bridges which after breakage led to loss of the W s gene. Somatic loss of Ws from the pbg chromosome was found to be responsible for chlorophyll variegation. A pbg chromosome which frequently formed mitotic anaphase bridges (more than 5 % ) was termed unstable in comparison to the stable one which formed less than 5% mitotic bridges ( GUPTA 1968). The investigation reported here revealed that even a chromosomal fragment of N . plumbaginifolia carrying the gene Ws, could cause chlorophyll variegation in tbc-pbg hybrid derivativles without forming somatic bridges.

Spontaneous morphological alterations of chromosomes in Nicotiana hybrids.
EVERAL workers have already reported that hybridization between Nicotiana species can bring about chromosome instability. One conspicuous case is loss of somatic plumbaginifolia chromosomes in derivatives of N . tabacum X N . plumbaginifolia hybrids (MOAV 1961). The present paper deals with another peculiarity of alien chromosomes in tobacco hybrid derivatives. It reports on heritable changes i...
متن کاملGene control of non-Mendelian variegation in Nicotiana tabacum.
I N the great majority of reports involving non-Mendelian plastid inheritance, the data indicate that nuclear genes exert no influence on the inheritance of abnormal plastids. These plastid mutations are assumed to arise spontaneously and to be irreversible. The inheritance of this type of non-Mendelian chlorophyll abnormality has been summarized in comprehensive reviews by RENNER (1936) and CO...
متن کاملEffect of radiation dosage on efficiency of chloroplast transfer by protoplast fusion in Nicotiana.
Chloroplasts of Nicotiana tabacum SR1 were transferred into Nicotiana plumbaginifolia by protoplast fusion. The protoplasts of the organelle donor were irradiated with different lethal doses using a (60)Co source, to facilitate the elimination of their nuclei from the fusion products. After fusion induction, clones derived from fusion products and containing streptomycin-resistant N. tabacum SR...
متن کاملInterspecific chloroplast recombination in a Nicotiana somatic hybrid.
Genetic recombination between chloroplasts of two flowering plant species, Nicotiana tabacum and Nicotiana plumbaginifolia, after somatic cell fusion is described. The parental lines differed in three cytoplasmic genetic markers. The N. tabacum mutant SR1-A15 was streptomycin-resistant, defective in chloroplast greening, and lincomycin-sensitive. The N. plumbaginifolia mutant LR400 was streptom...
متن کاملCloning and Expression of TNF Related Apoptosis Inducing Ligand in Nicotiana tabacum
Molecular farming has been considered as a secure and economical approach for production of biopharmaceuticals. Human TNF Related Apoptosis Inducing Ligand (TRAIL) as a promising biopharmaceutical candidate has been produced in different expression hosts. However, little attention has been paid to molecular farming of the TRAIL in spite of numerous advantages of plant expression systems. Theref...
